How are motions occurring on different timescales connected, and how do they contribute to catalytic efficiency? Have proteins evolved to preferentially access the conformational substates required for function? Recent advances in structural biology, protein engineering, and X-ray crystallography are providing new insights into these questions. Studies of directed evolution have revealed how enzyme dynamics can be optimized to produce highly efficient artificial catalysts, while developments in crystallographic data collection have enabled visualization of the molecular mechanisms underlying temperature adaptation in exceptionally proficient enzymes.
